From Dubrovnik , drive up the coastal highway until you reach the Ston exit, then turn into it it's about an hour from Dubrovnik. Keep driving until you reach Prapratno from the highway, Prapratno is about 10km drive. There you can get the car ferry: jadrolinija. I'm heading off to Mljet again on Sunday, but using the catamaran.

Highly recommended as a one day trip. In Korcula, ship's crew doesn't know if they are berthing in east or west harbor until 10 minutes before arrival! It is possible for ship to leave from different port than when it arrived from Dubrovnik. Ask in "Korkyra info" 15 min.
